== Ligation reaction == The mechanism of the ligation reaction was first elucidated in the laboratory of I. Robert Lehman. Two fragments of DNA may be joined by DNA ligase which catalyzes the formation of a phosphodiester bond between the 3'-hydroxyl group (-OH) at one end of a strand of DNA and the 5'-phosphate group (-PO4) of another. In animals and bacteriophages, ATP is used as the energy source for the ligation, while in bacteria, NAD+ is used. The DNA ligase first reacts with ATP or NAD+, forming a ligase-AMP intermediate with the AMP linked to the ε-amino group of lysine in the active site of the ligase via a phosphoramide bond. This adenylyl group is then transferred to the phosphate group at the 5' end of a DNA chain, forming a DNA-adenylate complex. Finally, a phosphodiester bond between the two DNA ends is formed via the nucleophilic attack of the 3'-hydroxyl at the end of a DNA strand on the activated 5′-phosphoryl group of another. A nick in the DNA (i.e. a break in one strand of a double-stranded DNA) can be repaired very efficiently by the ligase. However, a complicating feature of ligation conducted presents itself when ligating two separate DNA ends as the two ends need to come together before the ligation reaction can proceed. In the ligation reactions conducted in a laboratory, the ligation of DNA with sticky or cohesive ends, the protruding strands of DNA may be annealed together already, therefore it is a relatively efficient process as it is equivalent to repairing two nicks in the DNA.

==== Toronto City Council ==== The Ford government passed the Better Local Government Act into law on August 14, 2018. Premier Ford announced the controversial bill on September 27, on the last day for candidate registration for the October 22, 2018 Toronto municipal election, newly elected Premier of Ontario Doug Ford introduced the Better Local Government Act (Bill 5) which requires that Toronto use the same ridings for all its elections—municipal, provincial, and federal—effectively reducing the Toronto City Council from 47 seats to 25. Bill 5 passed on August 14, 2018. Toronto is Ontario's capital city and the largest city in Canada with a population of 2.7 million. The number of Council seats had just been expanded following approximately four years of consultations and debates. Ford said that these reductions would lower the cost to taxpayers by $CDN 25 ($USD19.1) million dollars in Toronto's $CDN11.1 ($USD8.5) billion dollar budget. Bill 5 reset the positions of regional municipality chairs as by appointment not be election in Peel, York, Niagara and Muskoka.
=== Complications === Pressure ulcers can trigger other ailments, cause considerable suffering, and can be expensive to treat. Some complications include autonomic dysreflexia, bladder distension, bone infection, pyarthrosis, sepsis, amyloidosis, anemia, urethral fistula, gangrene and very rarely malignant transformation (Marjolin's ulcer – secondary carcinomas in chronic wounds). Sores may recur if those with pressure ulcers do not follow recommended treatment or may instead develop seromas, hematomas, infections, or wound dehiscence. Paralyzed individuals are the most likely to have pressure sores recur. In some cases, complications from pressure sores can be life-threatening. The most common causes of fatality stem from kidney failure and amyloidosis. Pressure ulcers are also painful, with individuals of all ages and all stages of pressure ulcers reporting pain.

The meeting adopted a joint resolution by the Central Committee, State Affairs Commission, and the Cabinet, "on more thoroughly taking national measures for protecting the life and safety of our people from the worldwide epidemic disease". The resolution called to take strict national countermeasures to thoroughly check the inroads of the virus due to the steady spread of the epidemic. From mid to late April, restrictions on foreigners travelling in Pyongyang were relaxed, Nampo harbour was reopened to container ships, and the 14th Supreme People's Assembly with hundreds of delegates was held without the wearing of face masks. On 23 April, US analyst website 38 North reported that North Korea's early and extensive response appeared to be successful in containing the virus.
