Selank raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2026-01-12. Anything still debated is marked as such rather than presented as settled.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a structural analogue of tuftsin, a naturally occurring tetrapeptide fragment of the immunoglobulin heavy chain. The added Pro-Gly-Pro tail was intended to slow enzymatic degradation and extend biological activity. In Russia it is registered as an anxiolytic nasal preparation, while regulators elsewhere have not approved it for clinical use.
Proposed mechanisms centre on modulation of the GABAergic system, with reports of altered expression of genes related to GABA-A receptor subunits and changed monoamine turnover. Some studies describe inhibition of enkephalinase, the enzyme that degrades endogenous enkephalins, which may prolong opioid peptide signalling. Effects on brain-derived neurotrophic factor and on cytokine expression have also been reported. These findings come largely from animal models and small human studies, and the precise primary target remains unresolved.
Published clinical evidence is limited. Most controlled trials were conducted in Russia, enrolled modest numbers of participants, and appeared in Russian-language journals, which restricts independent verification. Reported outcomes include lower anxiety scores, improved attention and memory measures, and changes in fatigue ratings. Reviews written in English note methodological limitations such as small samples and inconsistent endpoints. Whether the compound produces clinically meaningful benefit relative to established anxiolytics is therefore an open question rather than an established finding.
Laboratory work relies on standard behavioral paradigms. Rodents are tested in the elevated plus maze, open field, and passive avoidance tasks, with outcomes compared against diazepam or vehicle controls. Intranasal dosing is used most often because it bypasses first-pass metabolism, though intraperitoneal and intravenous routes also appear in published protocols. Biochemical endpoints include tissue BDNF concentrations, cytokine levels, and monoamine metabolites. Human data are limited to small Russian trials reporting reduced anxiety scores; most were not prospectively registered, and few employed independent outcome assessment.
Measuring peptide exposure inside the brain is technically difficult. Selank is degraded rapidly in plasma, and assays must separate intact peptide from fragments, which favors targeted mass spectrometry over immunoassays alone. Reported half-lives are short, on the order of minutes, so effects observed hours later are attributed to downstream signaling rather than to the parent compound. Blood-brain barrier permeability is debated and rarely quantified directly. Gaps include absent dose-response characterization, inconsistent reporting of purity, and almost no pharmacokinetic data from human participants.
Selank is studied chiefly as an animal-model anxiolytic with proposed secondary effects on memory and immune signaling. Reported mechanisms include modulation of the GABA-A receptor complex, inhibition of enkephalin-degrading enzymes, and shifts in monoamine turnover within limbic structures. Some experiments describe increased expression of brain-derived neurotrophic factor in the hippocampus after repeated dosing. No single molecular target has been confirmed, and the peptide does not bind any receptor with the selectivity typical of a conventional small-molecule drug. Mechanism therefore remains a set of hypotheses rather than an established pathway.

Lyophilised material kept dry at minus 20 degrees Celsius or colder is the most stable form, and suppliers commonly state a shelf life of two years or more under those conditions. Once dissolved, degradation accelerates through hydrolysis and deamidation, particularly at alkaline pH or elevated temperature. Working solutions are usually divided into single-use aliquots to avoid repeated freeze-thaw cycles. The choice of reconstitution solvent affects both stability and the ionic strength of the final preparation.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres is the standard purity method. Mass spectrometry, typically electrospray ionisation, confirms identity through the expected mass-to-charge pattern. Amino acid analysis can verify composition independently. Chiral purity requires separate techniques such as derivatisation followed by chromatographic separation, and such data are rarely reported for research-grade material.
Quantification in biological matrices relies on liquid chromatography coupled to tandem mass spectrometry with stable-isotope internal standards. Low plasma concentrations and adsorption to container surfaces both complicate measurement. Solid-phase extraction is often needed to reduce matrix interference before injection. Reported limits of quantification differ widely between laboratories, which makes direct comparison of pharmacokinetic results difficult and limits meta-analysis.
Regulatory status differs sharply by region. Selank holds a Russian marketing authorization, where it is supplied mainly as nasal drops, while authorities elsewhere have not approved it for medical use. Material sold internationally is therefore usually labeled as a research chemical rather than a medicine. Peer-reviewed publications come predominantly from Russian laboratories, and sample sizes are generally small. Whether the compound produces comparable effects under independent, well-controlled replication remains an open question that the broader literature has not settled.
Selank is a synthetic heptapeptide developed in Russia as a structural analogue of tuftsin, a naturally occurring immunomodulatory tetrapeptide. Its sequence, Thr-Lys-Pro-Arg-Pro-Gly-Pro, keeps the tuftsin core at the N-terminus and appends a Pro-Gly-Pro tail. Researchers at the Institute of Molecular Genetics in Moscow synthesized the compound during the 1990s while searching for peptides with combined anxiolytic and immunomodulatory activity. The added tail was intended to resist enzymatic cleavage and prolong the molecule's presence in circulation.
=== Solvation effects === Solvation significantly affects the basicity of amines. N−H groups strongly interact with water, especially in ammonium ions. Consequently, the basicity of ammonia is enhanced by 1011 by solvation. The intrinsic basicity of amines, i.e. the situation where solvation is unimportant, has been evaluated in the gas phase. In the gas phase, amines exhibit the basicities predicted from the electron-releasing effects of the organic substituents. Thus tertiary amines are more basic than secondary amines, which are more basic than primary amines, and finally ammonia is least basic. The order of pKb values (basicities in water) does not follow this order. Similarly, aniline is more basic than ammonia in the gas phase, but ten thousand times less so in aqueous solution. In aprotic polar solvents such as DMSO, DMF, and acetonitrile the energy of solvation is not as high as in protic polar solvents like water and methanol. For this reason, the basicity of amines in these aprotic solvents is almost solely governed by the electronic effects.
Cyclodextrins were called "cellulosine" when first described by A. Villiers in 1891. Soon after, F. Schardinger identified the three naturally occurring cyclodextrins: α, β, and γ, referred to as "Schardinger sugars". For 25 years, between 1911 and 1935, Hans Pringsheim in Germany was the leading researcher in this area, demonstrating that cyclodextrins formed stable aqueous complexes with many other chemicals. By the mid-1970s, each of the natural cyclodextrins had been structurally and chemically characterized and many more complexes had been studied. Since the 1970s, extensive work has been conducted by Szejtli and others exploring encapsulation by cyclodextrins and their derivatives for industrial and pharmacologic applications. Among the processes used for complexation, the kneading process seems to be one of the best.
==== Negative selection ==== Negative selection removes thymocytes that are capable of strongly binding with "self" MHC molecules. Thymocytes that survive positive selection migrate towards the boundary of the cortex and medulla in the thymus. While in the medulla, they are again presented with a self-antigen presented on the MHC complex of medullary thymic epithelial cells (mTECs). mTECs must be Autoimmune regulator positive (AIRE+) to properly express tissue-specific antigens on their MHC class I peptides. Some mTECs are phagocytosed by thymic dendritic cells; this makes them AIRE− antigen presenting cells (APCs), allowing for presentation of self-antigens on MHC class II molecules (positively selected CD4+ cells must interact with these MHC class II molecules, thus APCs, which possess MHC class II, must be present for CD4+ T-cell negative selection). Thymocytes that interact too strongly with the self-antigen receive an apoptotic signal that leads to cell death. However, some of these cells are selected to become Treg cells. The remaining cells exit the thymus as mature naive T cells, also known as recent thymic emigrants. This process is an important component of central tolerance and serves to prevent the formation of self-reactive T cells that are capable of inducing autoimmune diseases in the host.

=== Placement of the pumpkin === Prior to October 8, 1997, an unknown individual or group of individuals placed a pumpkin on the lightning rod of McGraw Tower at Cornell University, 173 feet (53 m) in the air. The pumpkin was nearly 2 feet (61 cm) in diameter, and university spokesman David Brand would later state that the pumpkin was carved to fit the rod. They likely climbed up the tower's staircase which was open to the public at specified times, moved through the tower's bell cage without tripping the wires that would sound the chimes, cut a lock near the roof, emerged through a small service hatch at the bottom of the roof, and climbed 20 feet (6.1 m) up the steeply pitched steel roof of the tower to do so.
Despite guidelines recommending that intensive blood sugar control be based on balancing immediate harms with long-term benefits, many people – for example people with a life expectancy of less than nine years who will not benefit, are over-treated. It is recommended that all people with type 2 diabetes get regular eye examinations. There is moderate evidence suggesting that treating gum disease by scaling and root planing results in an improvement in blood sugar levels for people with diabetes.
=== Cosmetics === Some hair shampoos on the market include estrogens and placental extracts; others contain phytoestrogens. In 1998, there were case reports of four prepubescent African-American girls developing breasts after exposure to these shampoos. In 1993, the FDA determined that not all over-the-counter topically applied hormone-containing drug products for human use are generally recognized as safe and effective and are misbranded. An accompanying proposed rule deals with cosmetics, concluding that any use of natural estrogens in a cosmetic product makes the product an unapproved new drug and that any cosmetic using the term "hormone" in the text of its labeling or in its ingredient statement makes an implied drug claim, subjecting such a product to regulatory action. In addition to being considered misbranded drugs, products claiming to contain placental extract may also be deemed to be misbranded cosmetics if the extract has been prepared from placentas from which the hormones and other biologically active substances have been removed and the extracted substance consists principally of protein. The FDA recommends that this substance be identified by a name other than "placental extract" and describing its composition more accurately because consumers associate the name "placental extract" with a therapeutic use of some biological activity.

== Metabolism == cADPR and ADPR are synthesized from NAD+ by the bifunctional ectoenzymes of the CD38 family (also includes the GPI-anchored CD157 and the specific, monofunctional ADP ribosyl cyclase of the mollusc Aplysia). The same enzymes are also capable of hydrolyzing cADPR to ADPR. Catalysis proceeds via a covalently bound intermediate. The hydrolysis reaction is inhibited by ATP, and cADPR may accumulate. Synthesis and degradation of cADPR by enzymes of the CD38 family involve, respectively, the formation and the hydrolysis of the N1-glycosidic bond. In 2009, the first enzyme able to hydrolyze the phosphoanhydride linkage of cADPR, i.e. the one between the two phosphate groups, was reported. SARM1 and other TIR domain-containing proteins also catalyze the formation of cADPR from NAD+.

Firefighters hit the demonstrators with powerful water cannons, and the police continued to beat and arrest people. Protesters managed to build a defensible barricade in front of the Dunărea ("Danube") restaurant, which stood until after midnight, but was finally torn apart by government forces. Intense shooting continued until after 03:00, by which time the survivors had fled the streets. Records of the fighting that day include footage shot from helicopters that were sent to raid the area and record evidence for eventual reprisals, as well as by tourists in the high tower of the centrally located InterContinental Hotel, next to the National Theatre and across the street from the university. It is likely that in the early hours of 22 December that the Ceaușescus made their second mistake. Instead of fleeing the city under cover of night, they decided to wait until morning to leave. Ceaușescu must have thought that his desperate attempts to crush the protests had succeeded, because he apparently called another meeting for the next morning. However, before 07:00, his wife Elena received the news that large columns of workers from many industrial platforms (large communist-era factories or groups of factories concentrated into industrial zones) were heading towards the city centre of Bucharest to join the protests. The police barricades that were meant to block access to Piața Universității (University Square) and Palace Square proved useless. By 09:30 University Square was jammed with protesters.
Roy Rogers Franchise Company, LLC is an American chain of fast food restaurants headquartered in Ballenger Creek, Maryland, with a Frederick postal address, and primarily located in the Mid-Atlantic and Northeastern United States. The chain originated as the rebranding of the RoBee's House of Beef chain of Fort Wayne, Indiana, acquired by the Marriott Corporation in February 1968. However, Marriott first used the Roy Rogers Roast Beef name on conversions of the company's Junior Hot Shoppes in the Washington, D.C. area in April 1968, then the existing RoBee's stores. An aggressive nationwide franchising campaign was launched. At its peak, the chain included over 600 locations. The chain now has 42 locations in five states, either company owned or franchised. The Roy Rogers chain was sold in 1990 to Imasco, then the parent company of Hardee's, and experienced severe decline as many locations converted to Hardee's. In 2002, the trademark was purchased by Plamondon Companies. Roy Rogers' menu consists primarily of hamburgers, roast beef sandwiches, fried chicken, nine side items (including french fries), and beverages. Many locations also serve breakfast.
=== Analogues and derivatives === Analogues of 2C-B include 2C-I, 2C-C, DOB, 4C-B, 2C-B-FLY, and 25B-NBOMe, among others. DOB and 4C-B are α-alkyl derivatives of 2C-B, specifically the amphetamine (α-methyl) and phenylisobutylamine (α-ethyl) derivatives, respectively. β-Substituted derivatives of 2C-B such as BOB (β-methoxy-2C-B), BOH-2C-B (β-hydroxy-2C-B), βk-2C-B (β-keto-2C-B), and β-methyl-2C-B (BMB) have been described. βk-2C-B shows dramatically reduced potency and efficacy as a serotonin 5-HT2A receptor agonist compared to 2C-B. A variety of N-substituted derivatives of 2C-B have been tested, including N-methyl-2C-B, N,N-dimethyl-2C-B, N-ethyl-2C-B, and N-benzyl-2C-B. Most simple alkyl derivatives were considerably less potent than 2C-B, with N-ethyl-2C-B for instance having a 40 times lower affinity for the serotonin 5-HT2A receptor. The N-benzyl derivative however was found to have higher affinity than 2C-B itself, with N-(4-bromobenzyl)-2C-B binding even more tightly. This initial research did not include functional assays of activity, but later led to the development of potent substituted N-benzyl derivatives such as 25B-NBOMe, and 25B-NBOH. Another N-substituted derivative, 2C-B-AN, is an N-benzylphenethylamine-like prodrug of 2C-B. 2C-DB (6-bromo-2C-B) is a ring-substituted derivative of 2C-B. TWEETIO derivatives of 2C-B, in which one or both of the methoxy groups of 2C-B are extended to ethoxy groups, exist as well, such as 2CB-2-EtO.

It is a seven-amino-acid peptide, Thr-Lys-Pro-Arg-Pro-Gly-Pro, produced by chemical synthesis rather than extracted from biological tissue. Its design is based on tuftsin, a natural immunomodulatory tetrapeptide. The C-terminal Pro-Gly-Pro segment is a common stabilising motif in short regulatory peptides.
The core four residues correspond to tuftsin, which occurs naturally as part of immunoglobulin G. The full seven-residue sequence, however, is not a known endogenous peptide. It is a laboratory-designed analogue intended to combine tuftsin-like activity with greater resistance to breakdown.
No single receptor has been confirmed as the primary target. Reports describe involvement of the GABAergic system, interference with enkephalin degradation, and shifts in neurotrophic factor expression. Because these observations come from different models and assays, they have not yet been integrated into one accepted mechanism.
Intranasal administration predominates in both animal and human research because it avoids hepatic first-pass metabolism. Injectable and intraperitoneal routes appear in animal work mainly for comparison.
